One terminal group from Melanotan II
These two compounds are far closer than their separate names suggest, and understanding the relationship is the most useful thing on this page.
Melanotan II is an acetylated cyclic heptapeptide closed by a lactam bridge, terminating in a carboxamide. This compound is the same acetylated cyclic heptapeptide, closed by the same lactam bridge, terminating in a free carboxylic acid instead. That single terminal difference is the whole structural distinction, and it is not arbitrary: this compound arises as a metabolite of the amide form, which is how it was identified.
The consequence is a shift in receptor preference rather than a change of family. Both engage multiple melanocortin receptors. The amide form is described in the literature as the more promiscuous of the two, with relatively greater activity at the melanocortin-1 receptor found on melanocytes. The free acid is described as weighted more toward the melanocortin-4 receptor. Weighted more toward is not the same as selective for, and any experimental design treating this compound as an MC4R-only tool without controls for the other subtypes is assuming a cleanliness the pharmacology does not support.

Which is why identity confirmation is not a formality
Two compounds differing only in whether a terminal group is an amide or an acid differ in mass by approximately one dalton. On the free acid the protonated ion sits near 1026 and the doubly protonated ion near 513. The amide form sits one unit lower.
One dalton is a small separation but a real one on a competent instrument, and it is not something a purity percentage or a retention time resolves on its own. This is a case where a certificate publishing the mass confirmation does work that a certificate reporting only a percentage cannot. Look at the ion series on the certificate above before anything else.

Mass spectrometry cannot distinguish a D-amino acid from its L counterpart because they weigh the same. Confirming stereochemistry requires chiral analysis, which is not part of this panel or of any standard peptide panel.
